Propidium Iodide And Annexin V Staining

less than a minute read

Propidium iodide and Annexin V staining is a dual-color method used to distinguish viable, early apoptotic, late apoptotic, and necrotic cells. By combining Annexin V binding with propidium iodide (PI) membrane-permeability testing, researchers can rapidly analyze cell death mechanisms with flow cytometry or fluorescence microscopy.

Introduction to Propidium Iodide and Annexin V Staining

Cells do not all die in the same way, and the timing of membrane damage matters. In real terms, during early apoptosis, a cell may still maintain an intact plasma membrane even though it is actively undergoing programmed cell death. A conventional viability dye added too early may therefore miss these cells.

The Annexin V/PI assay addresses this limitation. Annexin V detects phosphatidylserine exposure on the outer leaflet of the plasma membrane, while PI identifies cells whose membranes have lost integrity. When the two stains are used together, researchers gain more information than they would from either stain alone.

This method is widely used to:

  • Measure apoptosis after drug, radiation, or gene-silencing treatments
  • Compare the cytotoxicity of experimental compounds
  • Assess immune-cell activation and cell death
  • Evaluate the effectiveness of cancer therapies
  • Monitor cell health during culture or manufacturing
  • Study injury, infection, and inflammatory responses

Although the assay is straightforward, its
